Dear Histonetters,
This is with regard to safranin-O & Toluidine
blue staining of rat bone cartilage. At last I got a perfect and beautiful
staining of cartilage. The procedure is as given by Gayle Callis and the
fixative and decalcification fluid are 10 % NBF & buffered formic acid
respectively. As our experts in histonet pointed out, that the real culprit
was EDTA used as decalcification fluid. Thanks to all for their sound
suggestions and prompt responses.
