[Histonet] Hale vs. Alcianblue

According to McManus & Mowry, Staining Methods (1960). page 137:
"Alcian blue is preferable to the Hale or Rinehart iron stains in that only one solution is required and direct staining occurs readily."  Their stain solution was 1% alcian blue in 4% acetic acid, pH 2.6.
Use only alcian blue powder from a batch certified (or recertified) by the Biological Stain Commission less than 5 years ago. This is one of the very few dyes that can deteriorate in the solid state. The chemical identities of dyes sold as "alcian blue" are variable; none are the original CI 74240, Ingrain blue 1. See Ali et al (2022) Biotech. Histochem. 97:11-20.

Dear all!

Has anyone informations, if the Hale-stain (colloidal iron or Mueller-Mowry)
can be substituted by Alcianblue-stain in any application?



We only use the Hale-stain for renal cellcarcinoma for differentiation
between chromophobe and clearcell carcinoma. And I’d like to know, if this
is really necessary.
