[Histonet] Manual double staining IHC

Hi Charles, protocols are going to depend on the species of your primary Abs you're looking to combine.  If it's mouse and rabbit you can stain simultaneously.  I use Biocare's double stain 1 or double stain 2 polymer depending on which marker you want on what colour.

If your Abs are the same species then you may need to stain sequentially with a denaturation step in between.

Aside from the terrible legal  liability that you would open your institution to by asking them to operate "dangerous" equipment when they are not in your employ, sitting at a strange microtome/waterbath set up with different knives, paraffins, processing, just would not be an accurate test.
I really liked Thomas Podawiltz's idea.  Usually I just ask leading questions and then evaluate during the probation period.

We have successfully done double staining using both DAB and Magenta on our Dako autostainer, you could do it manually.
