Jennifer,
You will not need to use the avidin/biotin block in your protocol. The polymer technology does not use any sort of biotinylation or avidin system for staining. That should help knock some time off your protocol.
You will need to make sure you use a peroxidase block (3% hydrogen peroxide) in your staining protocol to block endogenous peroxidase activity (namely red blood cells/sera).
