Mark,
Steve Ruzin has a book on plants, Plant Microtechnique and Microscopy,
which has a lot of info on wood from fixation to sectioning and staining
and including how to soften it up. Also there is another book - Plant Cell
Biology edited by Chris Hawes and Beatrice Satiat-Jeunemaitre, where you
might find some helpful information. I have never had the pleasure of
processing and cutting wood but I'm working on a project with seeds and
they are also very hard. I have just completed the processing and microtomy
awaits!
Good luck!
