Another cause could be partial lifting of parts of the section, allowing reagents to become trapped under the section. You commonly see it in thyroids where the colloid traps antibody and/or detection reagents under the colloid giving a particulate brown dust-like staining (sometimes looking like cocci). You can also sometimes see it in fibrous tissue in sections. Suggestions to solve this: check your sticky adhesive slides, or allow the slides to dry longer at room temperature before placing on the Bond (allows the adhesive time to work). I, too, was taught to cut the first level, then 3 unstained for possible IHC, then second and 3rd level. However, further research has shown that micrometastisis has little effect on patient outcomes so in many labs, the IHC performed on sentinel lymph nodes went out of "fashion". For those nodes that are frozen and show metastatic disease, there is little to gain from additional studies, other than one good H&E for permanent section. For other lymph nodes, we usually start with 3 levels for H&E, 40 microns apart, though I have worked before where the pathologists required 5 levels. I have noticed that the method I used using 1% BSA and 3% BSA blocking , then overnight with primary has reacted to my secondary goat anti Rabbit and mouse alexa dyre 594 and 488 with too much precipitation. Both the plasma cells AND stromal cells are stained in endometrial biopsies. The antibody is clone M115 and it is a predilute. The default protocol is ER1 for 30 minutes. I have tried to lessen the time in ER1 but it comes out the same. Leica support suggests adding a couple of washes after the Polymer step. Does anyone have any recommendations/thoughts?
