Not really because the Hematoxylin should "affect" only the nuclei UNLESS it is a nuclear antibody, in which case the antigenic reaction could be obscured. 
For nuclear Abs I used to dilute my hematoxylin as much as possible or did not use it at all.

Does a change in the type of hematoxylin counterstain require any re-validation of IHC stains?
