Hi Edie

Why risk a decolourisation step which may affect your final staining, there are recognized staining methods for Helicobactor using PAS such as Sayeed's method. It involves staining the gastric mucosa with PAS followed methylene blue giving blue organisms on a pink background. It is the same premise as the Alcian Yellow Toluidine blue procedure, so alternately toluidine blue could be used if that is your preference.

regards

We have a section that was incorrectly stained with PAS and no tissue left to recut. We need to do an H.Pylori stain on the slide. Any thoughts on how to destain the PAS? Tech tried acid alcohol to no avail.

Thanks
