Jennifer,

I used NaCl years ago in a clinical lab environment. We never put it in
antibody diluent. We did use it in our buffer rinses though, at a final
concentration of 0.9% (normal saline). It was thought to reduce
background staining. I don't know if it did in any great measure, but we
kept using it because it didn't hurt to have it in there. Since then we
have moved to automation and there is not any NaCl in our buffers.

Are you getting non-specific binding in your collagen with all your
antibodies, or just a couple?
