I think you are dehydrating your Von Kossa's correctly. After 20 to 30
sec in aqueous safranin O differentiate and dehydrate in 95% and 100%
alcohol. Then clear with 100% alcohol and xylene, then two changes of
As you know Von Kossa's demonstrates the presence of phosphates, soaps
and amorphous but not crystalline carbonates, rather than calcium
itself. Alcohol fixation is recommended as is neutral 10% formalin; any
fixative containing calcium must obviously be avoided.
