Make thin slices of bone and suspend in 10% soln of EDTA in 0.1M Tris buffer
pH 6.95 (add KOH pellets to achieve that). Keep soln in cold room (4 degrees
C) and stir with magnetic stirrer. Add bone which ought to decalcify within
24 hours. Freeze slices mount on chuck and cut sections; the knife may have
to be cooled with a block of CO2 ice or to cool the microtome.
