The intensity of DAB staining does not usually
indicate the quantity or concentration of an
antigen in an immunoperoxidase section. There 
are even circumstances in which the amount of oxidized 
DAB can vary inversely with the local concentration 
of the antigen. In general: enzyme amplification 
can detect and localize very small amounts of 
some antigens, but the intensity of staining has
no quantitative significance. 

There is a large body of peer-reviewed literature
in the field of quantitative immunohistochemistry.
Optical measurements correlate with antigen 
concentration only with the simplest methods.
