Greetings:
I have a question on behalf of a colleague. She is observing autofluorescence in thraustochytrids fixed in Davidson's AFA. The paper she is using as a reference for the acriflavine staining technique used 3.7% formaldehyde in seawater with Lugol's iodine (no concentration specified) as the fixative, and autofluorescence did not occur. Would anyone comment as to whether a difference in fixation would induce autofluorescence? Does Lugol's iodine quench autofluorescence?
Thanks for your insights.
Regards -
Carol
