[Histonet] LOST OF ANTIGENICITY

Hello everybody!

I do immunofluorescence with an AB against one aquaporine (AQP - water
channel) and mdrA (multi-drug resistance protein) on some fish tissues
fixed in paraformaldehyde or Bouin, embedded in paraffin and even sometimes
cut (1 or 3 years ago!!).
I can't manage to have any fluorescence even with very high concentrations
of AB. Do you think that it can be due to the age of the tissues? Can the
tissues loose the antigenicity when they have been embedded and/or cut for
such a long time? Does anybody have an idea of an other possibility?
Many thanks.

Anne-Sophie
