There are no specific regulations that require that it be separate. However, if you don't have a good training plan for new hires - or you have a team that isn't consistently dedicated to performing procedures by the book I have seen where IHC and other things like Neuro/Renal/Autopsy/Frozen sections are performed by only specific portions of an overall Histology team. The pitfalls of having dedicated IHC folks does mean if anyone is ever out on leave, or finds another position if you don't have enough people cross trained you can end up shooting yourself in the foot operationally. So I have found that trying to share the work across the team has worked best for me in my experience. I was the only person in the Immunopathology Laboratory (IP), which was started by a pathologist who did renal pathology.? At the same time, the Electron Microscopy Laboratory had three full-time employees.? I did the first immunoperoxidase stains (that's what we called them back then) in 1980.? As IHC became more popular, we rotated Histology personnel through the IP lab for 2 months at a time.? That did not work out well, since someone would not be back in the IP lab for at least a year or longer, and things changed monthly (new antibodies, new detection systems, etc.).? Our lab is still called IPOX, short for immunoperoxidase, so that tells you how long our lab has been around. We are a very large academic medical center and have 9 FTEs, 1 Supervisor, and 1 Technical Director (Ph.D) who works up all new antibodies and ISH probes. Our lab is directly adjacent to Histo and we need to be since Histo cuts/pulls all slides for IHC. Our IHC lab also performs DIF and Muscle biopsies on the day shift and we have a subset of our IHC techs that are trained on creating TMAs for our on-slide controls using the TMA Grand Master. Xylene-free mounting media may also be worth evaluating for laboratories seeking to reduce or eliminate xylene from their mounting-media workflow. As with any mounting medium, performance can depend on the specimen, staining procedure, mounting technique, coverslip, and characteristics of the UV light source. I have seen them removed with a hotplate and a razorblade. However, the tissue did not survive well and then had to be soaked in acetone and xylene. Seemed like a very permanent coverslip that would outlast storage. I would hope that anyone using the UV cured stuff would perfect their method prior to using patient tissue.
