The simplest response may prefer using a conjugated secondary if you're planning to do multiplex or multicolor analysis. If I was limited in commercially available options for primaries needed for the test sample, then i would prefer a conjugated secondary as it loosens up testing requirements for testing. Another benefit is that it may increase specific testing and allow positive staining to appear more visibly present than a directly conjugated antibody. There are other benefits for a conjugated secondary or tertiary antibody, but it requires users to know how to troubleshoot the stain. Direct conjugation is easier to use in many circumstances. I would communicate to your friend to try both for the study and determine as a team of internal collaborators on which method is easiest and adequate enough for performing the method testing. We have started to work on paraffin processing on E18.5 to P1/2 mouse embryos and would love to have insights into the issues we are having. We believe we are running into some processing issues as some of the organs such as the liver and heart are very crunchy, and they are not staying where they should be located. We have been poking some holes with needles in the tissue, but it seems not helping much. Would cutting into the skin be a better option? The protocol we have at the moment is: Tissue fixing in 4%PFA at 4 degree C overnight PBS washes 30mins 4 times at 4 degree C 30% Ethanol 2 hours 50% Ethanol 2 hours 70% Ethanol 2 hours And then subject to paraffin embedding machine for a long processing protocol that takes 2 days. I've seen protocols that use NBF at higher temperatures for fixation, however we are trying to do some spatial assays and RNA degradation is a concern if using a higher temperature. For our other tissues of bone and cartilage, we've used zinc buffered formalin at RT without issues downstream for spatial/FISH. I recently started a position with the University of North Texas Health Science Center as their Histology Research Associate. During a quick meeting with my director he mentioned that he was interested in possibly setting up a 'microcredentialing' program for Histology since our lab is now up and running. Pretty much from what he was telling me it would be similar to when we were in high school taking those computer classes where we would get the Microsoft certifications showing we knew how to use Word, Excel, Powerpoint, ect. My hope would be to try setting it up in such a way that students when finished would have a better understanding of what we do in Histology but also have the basics to possibly sit (depending on how long out program would last) for the HT exam using the route 2 option ["Both require prior education in the histology field, either through attending a histology program, or acquiring laboratory experience in the field."] My question for the field is 2 fold. 1. Is this something that you think could be a second path for people to get into the field of Histology due to our nation wide shortage? We deal with med students, pre med, medical research students, and starting a nursing program here next year. 2. If this could work what do you think I should focus on?
