Use frozen or paraffin sections, according to instructions that come with the primary antibody. For histochemical demonstration of acetylcholinesterase activity (seen also in some neurons that are not cholinergic), you need frozen sections of tissue fixed for 12h in neutral formaldehyde at 4oC. My favourite method for AChE activity was that of Karnovsky & Roots 1964 J. Histochem. Cytochem. 12:219-221. The reason we do this rather than wax histology is that our "lab" travels to some pretty remote areas (Papua New Guinea, Micronesia, etc) and you need less stuff to do resin histology (really only a JB-4 Kit, microtome, and some glass knives plus whichever staining method you prefer) and can even do it without power. We actually have a workshop coming up soon in Guam, if you are interested. Message-ID: Hello Histonet, I am using an old, but good, tissue processor (Sakura Tissue Tek II)?the plastic timing pins (photo attached) are old and the plastic is starting to break. I?ve tried to find replacement pins but have been striking out. I?m hoping that someone might have one of these tissue processors sitting around in storage and might be willing to send me a few extra timing pins. Or if anyone knows where I might be able to purchase some, I?d be happy to contact them (have checked ebay and a few tissue-processor re-furbishing businesses without luck).
