But are you asking if it needs special ventilation? If the bottle is sealed or has a well-sealing cap, then storing small quantities in the general lab is ok, so maybe a gallon or, better yet, several 500ml bottles. When we store in the general lab we keep the bottles as small as possible to prevent a large spill. However, if you have many gallons then, even if sealed, they should be stored in a well-ventilated area that is away from people. Either a dedicated chemical store room, or a chemical cabinet that is ventilated to the same ventilation system as the fume hoods. That is to prevent overwhelming fumes if one or more bottles should leak. It shows your interest & you will get to know your pathologists. This will also encourage them to feel more comfortable in the lab. Try different lab settings. Academic research you have a lot of interaction with scientific people & you will have to figure things out/research them yourself. Clinical is where most people start out & what Tim said is spot on. Pharma is really incredible, especially now. It will most likely mean working with animal, cell culture & archival materials. You?ll need to bring your A game to work each day, but most of the people in all these settings will be willing to show an eager learner. Good days come with bad days. I might have added to take every opportunity to do something different. While I started in a clinical setting I also worked in a federal government clinical/research setting and also for a vendor in the field. They both added immensely to understanding the breadth of the field. Especially the vendor work which was in product development and entailed visits to many labs and other vendors, even internationally. That work really opened my eyes to both sides of the field. If something like that comes along, and fits with family/life, it is well worth doing. Does anyone out there use this unit? It seems to be very popular with the research community. Do you ever adjust the temperature of the cutting head down that low, or do you just adjust the chamber temperature? Why would you ever use this separate feature? Everything gets very brittle at that temperature, including your hands after a short while. Never -60 though! Maybe that is just for the peltier freezing mechanism? For putting the tissue on the chucks with OCT glue? I typically will set the specimen head a little lower than the chamber. I don't like the roll plate on the Leica. The Leica design creeps... a lot. Also, with the leica, the contact point with the blade is a soft aluminum arm. If you make adjustments with the roll plate holder contacting the blade, it will damage the soft aluminum holder. I like the thermo design much better. It creeps a lot less. Here's a procedure for roll plate adjustment. 1. Mount block on specimen disc. 2. Place Specimen Disc in chuck, position Blade carrier, and face off. 3. Turn knurling screw clockwise until sections do not pass under the Anti-roll plate. 4. Turn knurling screw counter-clockwise by 20 degree increments until sections effortlessly pass under the Anti-Roll Plate WITHOUT hitting the block. * If the block is hitting the anti-roll plate, it may cause damage to glass. * Hard contact is visible and can be heard. * Soft contact, which can also damage the Anti-roll plate, cannot be seen or heard. But, can be felt by gently placing finger on the anti-roll plate turning lever while taking a section. I don't like the roll plate on the Leica. The Leica design creeps... a lot. Also, with the leica, the contact point with the blade is a soft aluminum arm. If you make adjustments with the roll plate holder contacting the blade, it will damage the soft aluminum holder. I like the thermo design much better. It creeps a lot less. Here's a procedure for roll plate adjustment. 1. Mount block on specimen disc. 2. Place Specimen Disc in chuck, position Blade carrier, and face off. 3. Turn knurling screw clockwise until sections do not pass under the Anti-roll plate. 4. Turn knurling screw counter-clockwise by 20 degree increments until sections effortlessly pass under the Anti-Roll Plate WITHOUT hitting the block. * If the block is hitting the anti-roll plate, it may cause damage to glass. * Hard contact is visible and can be heard. * Soft contact, which can also damage the Anti-roll plate, cannot be seen or heard. But, can be felt by gently placing finger on the anti-roll plate turning lever while taking a section. It creates aerosolized particles that can linger, and infect, for hours. At our lab, we long ago decided to forbid the use, and we have no problems cutting beautiful frozen sections without it.
