PRINCIPAL DUTIES AND RESPONSIBILITIES: Research in the Endocrine Unit focuses on metabolic bone diseases and mineral metabolism and ranges from trials of novel therapies for osteoporosis to use of genetically altered mice and cell biology to understand the factors that influence calcium and phosphate metabolism. Do you use the same EM-400 paraffin for processing? If you use different paraffins that might contribute to your sectioning problems. If the waxes are different the tissue and the rest of the block are not one continuous unit in the "eyes" of the knife. The waxes have different properties so they cut differently because the embedding wax does not infiltrate into the tissue but wraps around it like a blanket. I'm not sure if that's what your lab does but if it is I suggest using the same paraffin for processing and embedding. At one time, our cassette printers (Leica IPC) were not interfaced either, but we were able to use Leica's software and enter the cassette identifiers, and that information was able to be saved in the bar-code, which also printed on the cassette. Patient names were not printed, only the accession number/letter. We have the SlideMate Pro printers, and before they were interfaced, we were able to scan the barcode on our blocks and they would print the information obtained from there. The SlideMates were also able to be programmed so that the hospital name was printed at the bottom of each slide. So it can be done, but only if your blocks are bar-coded.
