But it's been 20+ years since the last one I bought, and I have no clue what features I'll need now. Do I buy a reliable 90s analog model? Do I need digital control? No idea! I'd love to talk to sales reps to get the pitch...but because I'm most likely to buy a used model, I feel like I'd be wasting their time. However more importantly I anticipate a lot of IHC/IF, and probably most important to us is preserving lipids in sectioned tissue (hence avoiding the clearing agents used with paraffin). > "What kind of tissue?" We're a tissue engineering company working extensively with mammary cells, so the workload will likely be fatty tissue, and some composite samples that are tissue and polymer scaffolds (nothing a good blade can't easily cut--I think). > "What volume?" Pretty low. I may well be underestimating the workload, but most likely a dozen blocks every few weeks. > "What budget?" That what I need help with. I can probably go up to $20k, and mgmt is OK with a new model if it's necessary, but frankly I wasn't fantastic on a cryostat the last time I bought one so I'm not confident I'm qualified to say what exactly we need. > "What is your skill level?" Noob. I've used a cryostat before, but my technique...definitely needs improvement. On the other hand, I'm the only one in my company that has any experience with histology at all, so I anticipate training a new generation of research histologists. So, I probably want a cryostat that may not do everything for me, but also won't require a lot of shoehorning. > > I'll also leave a more open question down here: What is a paraffin microtome better suited to than a cryostat, and vice-versa? > > Thank you so, so much in advance for your time and any help you can offer!!! > > Warmest regards and aloha, > Jack England > > > ------------------------------ > > Subject: Digest Footer > > _______________________________________________ > Histonet mailing list > Histonet at lists.utsouthwestern.edu > http://lists.utsouthwestern.edu/mailman/listinfo/histonet > > ------------------------------ > > End of Histonet Digest, Vol 228, Issue 13 > ***************************************** From lesraff at gmail.com Tue Dec 6 07:41:07 2022 From: lesraff at gmail.com (Lester Raff) Date: Tue, 6 Dec 2022 07:41:07 -0600 Subject: [Histonet] One more month until it's over--why it is time to re-tire Message-ID: https://raffstuff.home.blog/2022/12/06/the-tri-state-trips-me-up-one-last-time/ Les Raff From denise.long at uconn.edu Tue Dec 6 13:56:35 2022 From: denise.long at uconn.edu (Long, Denise M.) Probably on Friday June 9th. There will be invited presentations and poster sessions. Details are not yet available but should appear on the Commission's web site at some time in the next few weeks. Keep an eye on https://biostain.com. On the BSC web site you can also see what was presented at about 20 earlier BSC meetings (at https://biologicalstaincommission.org/category/annual-meetings/). The BSC web site also has many other links useful to histotechnologists, notably a Glossary and a variety of What's New? items, with conspicuous links on the Home page. The glossary comes up as the top item if you type stain glossary (or similar) into Google. I can see, Lisa, that you won't be travelling from Australia to the USA for a one-day meeting! Your Histonet query has provided an opportunity to advertise the BSC's forthcoming centennial celebration. The availability of BSC-Certified stains (about 60 dyes, as powders) underlies the validity of much medical and other biological research and of all routine staining done in pathology labs with bought pre-made dye solutions. Beware of fake "certified" or similarly named stains sold perhaps from China and India and marketed in Europe and the Americas. These do not carry the BSC's small and difficult-to-forge bottle label, which subtly indicates the history of every truly certified stain. They are absolute workhorses; printing is fast and beautiful and the maintenance is really easy. The only downsides I can think of is that the IPC has a large footprint, and the ink is really pricey. You're supposed to change the cartridge every 3.5 months or 60,000 prints or so. We never hit that volume so we're always throwing away ink! Thanks, Brian D.
