Shortening the longer schedule produced underprocessed fatty tissues and over processed biopsies, so we run the biopsies on the short schedule overnight, run a quick clean cycle, then the larger tissues on the 10hr schedule on the same instrument, pull the them off and leave them in the embedding unit to embed in the afternoon, or next morning. It does put the larger tissue a day behind, but does not sacrifice the quality of the processing. It you shorten your large tissue processing run so that you can run small biopsies with the bigger stuff, you will have to revalidate your IHC, so maybe this might be a better alternative. One in the am and one in the pm. Then run your large blocks over night as per usual. Any late arriving small biopsies can be run on the am run the following day . This way everything is processed within 24 hours of arrival maintaining your TATs. Message-ID: Dear List, I was cryo-sectioning fixed-frozen and fresh-frozen mouse brains, and found a lot of wrinkles and folding in the fixed-frozen sections, while the fresh-frozen sections were pretty smooth. Below are my sectioning conditions and observations. I would appreciate any advice on improving the fixed-frozen sections. Tissues: fixed-frozen mouse brains (OCT embedded), fresh-frozen mouse brains (OCT embedded). Tissue temperature: at -80 deg for at least 48hrs, equilibrated to -18 deg for 1hr before sectioning. Thickness: 14um Slide: Superfrost plus Cryostat chamber and head temperatures: -18 deg, -21 deg respectively. Observations: When the fresh-frozen sections came out of the block, they rolled up smoothly, ?jumped? to the slide immediately, and unrolled themselves smoothly. But the fixed-frozen sections looked more ?limp? ? they were ?wavy?, could not roll up, and when I put a slide over them, they could not extend smoothly thereby creating wrinkles, folding and even bubbles. I have attached an image showing my fresh-frozen sections at the top, and fixed-frozen at the bottom: https://drive.google.com/file/d/1FMWcW7g9N_yUrhsiEMe8orvR3vM32RiU/view?usp=sharing It looked as if the fixed-frozen block was ?warmer? than the fresh-frozen block, but in fact they had been equilibrated to -18 deg for the same amount of time (~1hr). And I believe it was also not a blade artifact, because I alternated between cutting fixed-frozen and fresh-frozen sections several times using the same blade. The fresh-frozen sections looked fine each time, but the fixed-frozen sections were always problematic. I also believe there was no temperature difference between the slides used for fixed- or fresh-frozen sections (both were stored at room temperature). My protocol for preparing the fixed-frozen tissues is as follows: 1. Transcardial perfusion of PBS (10ml) and 4% PFA (10ml). 2. Fixation with 4% PFA at 4?C overnight (~18hrs). 3. Wash with PBS (20min x 3). 4. Dehydration with 15% sucrose (in 1X PBS) at 4?C until tissue sinks (~12hrs). 5. Dehydration with 30% sucrose (in 1X PBS) at 4?C until tissue sinks (~24hrs). 6. Embedding in OCT with dry ice and 100% EtOH until OCT solidifies. 7. Stored at -80?C. What do you think I could do to improve the fixed-frozen sections? Your input would be much appreciated. Message-ID: Dear List, I was cryo-sectioning fixed-frozen and fresh-frozen mouse brains, and found a lot of wrinkles and folding in the fixed-frozen sections, while the fresh-frozen sections were pretty smooth. Below are my sectioning conditions and observations. I would appreciate any advice on improving the fixed-frozen sections. Tissues: fixed-frozen mouse brains (OCT embedded), fresh-frozen mouse brains (OCT embedded). Tissue temperature: at -80 deg for at least 48hrs, equilibrated to -18 deg for 1hr before sectioning. Thickness: 14um Slide: Superfrost plus Cryostat chamber and head temperatures: -18 deg, -21 deg respectively. Observations: When the fresh-frozen sections came out of the block, they rolled up smoothly, ?jumped? to the slide immediately, and unrolled themselves smoothly. But the fixed-frozen sections looked more ?limp? ? they were ?wavy?, could not roll up, and when I put a slide over them, they could not extend smoothly thereby creating wrinkles, folding and even bubbles. It looked as if the fixed-frozen block was ?warmer? than the fresh-frozen block, but in fact they had been equilibrated to -18 deg for the same amount of time (~1hr). And I believe it was also not a blade artifact, because I alternated between cutting fixed-frozen and fresh-frozen sections several times using the same blade. The fresh-frozen sections looked fine each time, but the fixed-frozen sections were always problematic. I also believe there was no temperature difference between the slides used for fixed- or fresh-frozen sections (both were stored at room temperature). My protocol for preparing the fixed-frozen tissues is as follows: 1. Transcardial perfusion of PBS (10ml) and 4% PFA (10ml). 2. Fixation with 4% PFA at 4?C overnight (~18hrs). 3. Wash with PBS (20min x 3). 4. Dehydration with 15% sucrose (in 1X PBS) at 4?C until tissue sinks (~12hrs). 5. Dehydration with 30% sucrose (in 1X PBS) at 4?C until tissue sinks (~24hrs). 6. Embedding in OCT with dry ice and 100% EtOH until OCT solidifies. 7. Stored at -80?C. What do you think I could do to improve the fixed-frozen sections? Your input would be much appreciated. The cyto staff were also separate but were trained in some of the histology tasks (loading processors and accessioning). They could help out with those tasks if the histology lab was short staffed. I believe it would basically be the same as validating a new microtome except specimens (ie blocks) are plentiful for microtomy whereas for us, fresh tissue for cryosectioning is not readily available. Background: We are an acute care hospital and we only get about 50 intraoperative consult requests per year. Our older unit is still functional so I can do side-by-side comparisons with the new unit. Our frozens are only stained for H&E and a very rare (one every couple of years) Oil Red O. So I have two questions: 1. Can I use formalin-fixed tissue soaked overnight in a saturated sucrose solution for the validation? This seems reasonable since I can do side-by-side with the older unit. {autopsy tissue could be an option if the timing coincided with our validation but Murphy's Law will probably prevent this option! lol} 2. How many tissues would be satisfactory? Is one specimen of each of our typical frozen specimen types enough? We typically get: ovary, thyroid, skin and lymph node. He swore me to lifelong secrecy (which I'll have to honor, since I've forgotten the patient's name). The patient was a recently retired, quite prominent local businessman, whose transgender status was a deeply guarded secret. He'd been taking androgenic hormones for many years. I'd think a rather small number would suffice, perhaps four per breast. Cancer risk in such a patient should be low, because of hormonal suppression. About fifty years ago, when I was a resident at a large university hospital, I received bilateral simple mastectomy specimens from an older individual. On dissection the rather small amount of rather fibrous breast tissue was filled with numerous 5 to 10 mm cysts, all about the same size. (I've never seen anything like it, before or since.) I took some sections at random, having no idea what was going on, and the slides were quite unremarkable. I took the case to the senior pathologist, who was considered a major authority on breast pathology, to sign out. He swore me to lifelong secrecy (which I'll have to honor, since I've forgotten the patient's name). The patient was a recently retired, quite prominent local businessman, whose transgender status was a deeply guarded secret. He'd been taking androgenic hormones for many years. After his retirement, he had the long-put-off mastectomies performed. Yes, such things went on fifty years ago. I am not sure exactly what he wants and am not sure if this is possible. I have been asked to assist a researcher in examining thin sections of coagulated blood drops in order to distniguish individual drops that were deposited at different times onto a substrate. The investigator intends to deposit one drop of fresh blood onto a surface (perhaps paper or paraffin) and allow the blood to coagulate. After that is coagulated he will deposit a second drop on top of the first. Once the second drop is also coagulated he will preserve the sample as recommended for cross sectioning and evalaution. He intends to cut cross sections of the samples and examine them in order to determine how coagulation time changes the interface of the two droplets. The underlying question is "does time/extent of coagulation of deposited blood affect the deposition pattern of additional layers of blood on top of the first layer?" If the entire sample was encased in paraffin would there be a way to cut sections for microscopy, either with or without staining?
