Of course for IHC that means a revalidation/reverification of our IHC stains. Since I have not had to do this before I wanted to get some guidance on how to go about doing this. I feel like I have to completely revalidate the ER, PR and Her2 (20 positive, 20 negative) but wasn't sure about the other antibodies. We have a menu of around 130....do I have to test every one of them or can we chose a representative sample? How many antibodies would you suggest? How many positive and negative cases of each should we run? It says it is left up to the medical director but with our CAP inspection next spring we want to make sure we will be fully prepared. We deal with mostly mouse tissues and sometime samples will process over the weekend and come off on a day that no one can embed straight away. Samples are taken out of the processor and let sit on the bench and the paraffin coated cassette tissues harden. This delay can be hours or a few days if it's a weekend/ holiday. The samples are then placed in molten paraffin to melt and are embedded. We have done this with minimal problems but are wondering if some tissues like skin mouse ears could be more difficult due to this delay. Once the delay was 6 months. I allowed the cassettes with tissue in to solidify ( as do you) and then leave them in a labelled box to await embedding. The Leica instrument feels a bit more modern and the reagent management system is useful for reagent changing schedules. The changing of reagents and wax is pretty easy with the drainage hose. I've rarely had any issues with them, but have had plenty of issues with the Peloris instruments (made by Leica). The VIP is a workhorse. Very rarely have we had problems with the instrument. It's very reliable, but not quite as "flashy" as the Leica instrument. I'm not sure about parts for the VIP, but if you don't have a service plan for the ASP300S, be prepared to pay out the nose for parts. I was quoted over $1,000 for a wax drain hose that was literally a piece of plastic piping with a simple connector on it. I had a sister lab that got a new/used ASP300S and the hard drive died within a couple months. I would really recommend against them. The only Leica certified repair people in the Phoenix area would not work on my ASP300 because they break so often and they didn't want to be responsible for it. We recently acquired the VIP 6 AI to back up the remaining years of our VIP 5. The last time our VIP 5 had a PM it was down for 4 days which included a weekend and we only have one processor. The VIP 6 has many up grades for Solution Management. We have not had it long enough to give feedback. Sakura only makes the VIP 6 now so you may want to consider the VIP 6 instead of the VIP 5. Aqueous solutions tend to stain poorly. Sections are firstly stained with Celestine blue, followed by a regressive Harris's haematoxylin (excess stain is removed from the plastic with acid alcohol). The sections are then counterstained with buffered eosin Y/Phloxine B. Solutions 1. Celestine Blue - Iron Alum 2. Harris's Haematoxylin 3. Buffer for Eosin Y/Phloxine B. Earlier this month we had two slide drawers replaced. Yesterday one of them opened during a run and would not close again. Fortunately we were able to salvage the slide that was being stained in the drawer. A little later I was removing the remaining few slides from the module and noticed wisps of smoke coming from the drawer. I put the module in sleep mode and the drawer still would not close so I turned off the power. As soon as I did this the drawer closed. I phoned Roche and at their direction I turned the module on again and the drawer opened again immediately. It would not go into ready mode so we tried operating the compressor manually but to no avail. The drawer then made a crackling noise and smoke started coming out again. Has anybody else had a similar experience??? I'm surprised they told you to keep using it since it poses a real safety risk. If I can find a copy of the notice I will send it to you this week. We have 2 Discovery Ultras and 2 Discovery XT's. We had a drawer leaking onto electrical wiring on one of the XTs this past week. They "fixed" the issue a few weeks ago only for it to occur again, so this past week they installed a brand new drawer. Roche's lack of transparency is a real problem in my opinion. Thanks, Taylor C. I decided to do a std Pwax processing schedule. The student dissected the appropriate area then placed in 10% Formalin in PBS pH7.4 for 24hrs on a rocker ( gentle agitation) I then processed to Pwax using a std IMS/Xylene/wax on a Leica carousel ( 1hr in each station) Worked fine. Could do tinctorial and IHC no problem. Sure, many primary Abs didn't work....... We also did TUNEL on them Pwax sections....worked a treat. I may have TUNEL images in the Histonet Image archive. Good luck! NB: Sure, maybe freezing is a better way forwards. Depends. We have demoed a Sakura and Leica HistoCore BioCut but would like to test out at least 1/2 more options before deciding on where to proceed. Does anyone have any suggestions on microtomes that are good for both experienced tech and new graduates? We are also specifically searching for microtomes that have the capability to reverse the advance wheel directions (i.e spinning away from the body moves the block holder backwards but can be changed to do the reverse back on tech preference)
