I was wondering if anyone could provide me with a reliable H&E protocol for fresh frozen tissue. Currently, we cut our slides on the cryostat and store them at either -20 or -80 (depending on the study director) until they require us to stain for morphology. Typically the tissue isn't always perfectly snap frozen, and often times there are freezing artifacts. However, I would like to preserve as much as the morphology as possible without further damaging the tissue. I have really only had experience staining perfusion-fixed tissue using the following staining procedure: Water rinse Hematoxylin (7211 Richard Allen Scientific) for 30 seconds Water rinse under running tap Clarifier 1 (Richard Allen Scientific) with three quick dips Water rinse under running tap Bluing reagent (Richard Allen Scientfic) for 1 minute Water rinse under running tap 95% EtOH for 30 seconds Eosin-Y with Phyloxine (Richard Allen Scientific) for two quick dips 95% EtOH for 30 seconds 100% EtOH for 1 minute 100% EtOH for 1 minute 3, one-minute exchanges in Clear Rite 3 (a xylene substitute, Richard Allen Scientific) However, today I was asked to run an H&E on a fresh frozen slide. I post fixed in 4% PFA for 10 minutes immediately (not allowing the slide to thaw), and then ran the above procedure as normal. When I examined the slides under the scope, there appeared to be terrible freezing artifacts. The study director happened to think it was both a post-fix and staining issue, so I re-ran the procedure without post-fixing, (per his recommendation) as follows: Immediately into hematoxylin for 30 seconds Water rinse under running tap Eosin for 2 quick dips 95% EtOH for 1 minute 100% EtOH for 1 minute 100% EtOH for 1 minute 3, one-minute exchanges in Clear Rite 3 I saw slightly better morphology, but the director still was not happy with the outcome. I have spent the remainder of my afternoon researching different H&E protocols for fresh frozen tissue, and all seem to be relatively similar to the two mentioned prior. Does anyone out there in Histo-land consistently run H&E on fresh frozen tissue, specifically on slides that have been stored for a while? I work particularly with brain and spinal cord, but we occasionally will cut other tissue and organs. I would like to optimize this procedure as soon as possible. Thank you in advance! A negative would have you apply the stain to tissue that could not be stained by the stain, or not apply the specific stain to show there is no interference from other reagents used. The question is whether that is useful for tinctorial stains. The solution to these is to put some verbiage in your procedure that this does not require a negative control and why. You should have clear description, and pictures of what an acceptable stain looks like, and even more important, what a failed stain looks like. However, a negative should be used for organism stains, and could be useful for some specific tissue elements. For example, congo red or iron, you can leave off the specific stain and just use the counter stain (it could be useful - if the control is cut thru for instance the positive and negative would look the same). I know well that this is not the normal practice going back many decades, and we argued with the Joint Commission about this as well, but regulatory agencies are becoming stricter about validations and controls, so it could be a good thing to prove your systems work as intended and to catch the rare failure. The ideal solution would be to find another lab that does have this validated and let them do it. Your lab could validate it and have it ready for the next time, but it would probably be a rare procedure and many of the reagents would expire between uses. We are cutting sections with epidermis dermis and fat. We need to tease out all the folds and wrinkles but since the epidermis is separating it makes teasing the tissue folds and wrinkles out very difficult. Sometimes the tissue wafer will separate completely from the surrounding oct. If an embalmed body is to be autopsied, only the arterial embalming should be done first. If they do the "cavity embalming" as well they make a mess of the organs. We place both positive and negative controls on every IHC. Minimal design is one positive and one negative control. To start make a simple one with positive and negative using a 2mm skin punch biopsy with plunger to cut out known tissue and mount together. If you can't punch out tissue, or a valuable control is thin, then one can still make a miniature array. Make an accordion or fan by cutting a 50 micron section and scrunch together. Repeat with negative. Repeat with another positive. Wrap the whole thing in parafilm and Mount your layers on edge in paraffin. For example HHV8, KS marker, we have 5 controls 2 cases of KS, PG, hemangioma and normal skin. Steve A. If you have any advice I would greatly appreciate it! **************************************************************************** ********************************* Several things to think about: FF tonsil should be cryoprotected with 20 -30% sucrose before snap freezing to prevent ice crystal formation. Simply take the FF tonsil, cut into smaller pieces and immerse into 30% sucrose over night at 4C. When the tissue sinks to the bottom of this somewhat thick syrupy solution as which indicates the tissue is cryoprotected. Blot off excess sucrose, embed in OCT and snap freeze. You can snap freeze many blocks to stockpile controls and store these at -80C, shorter time at -27C. One thing you did not mention is: are tissues you are trying to do DIF on for diagnosis also formalin fixed? I would think a tissue control should be handled the same way as the test tissue i.e. fresh tissue frozen sections fixed with cold acetone compared to FF tissue. If your tissues are not FF, then why not collect fresh tonsil and snap freeze that as a control? You can make several blocks and store a -80C or even -27C (for a short time). The concentration of your antibodies may be different on a FF tonsil section due to the cross linking as compared to the concentration on a fresh tissue frozen section fixed with cold acetone. Antibody dilution would have to be tested. Personally, I would want my antibody concentration for a control and test tissue to be the same. If you already run DIF on a FF tissue from patient, then a FF control works fine. If you are using FF tonsil sections as controls, then FF tissue will have aldehyde induced autofluorescence but this can be removed. Autofluorescence is annoying when trying to read the sections and could mask what you want to see unless you use a contrasting color fluorophore. To get sections to stick, mount frozen section on reliable plus charge slides, and air dry the frozen section a bit but be gentle when rinsing the sections. Formalin fixed frozen sections always have the possibility of coming off, but many have success retaining the section and discussed many times on Histonet. There may even be newer plus charge slides touted for FF fixed frozen sections these days. Good luck Gayle M. I was wondering if anyone else has experienced a dramatic dropoff in staining signal with this antibody after a few months. I have now had this happen to me twice in the last 3 months. It stains very nicely when the vial is first opened and seems to hold staining intensity for about 3 months and then drops off dramatically even though the expiration date is 2 years out. I am using it on the Leica Bond. I have been in contact with CellMarque about this problem but they tell me they have not had any complaints from any other labs. I use many antibodies from CellMarque and have only seen this problem occur with one other antibody (ALK-1) about a year ago. Found out that my detection kits were from bad lots. Leica rep finally was able to swing by and helped me troubleshoot the issue. Found out there were 3 lots of bond polymer detection kits that were manufactured differently tan normal and it was affecting staining. The lot #'s ended in 555, 314, and 486. Lately I have been having trouble with our SDH staining too light. I've been working with two different protocols and have kept the slides in the SDH incubation solution from 1 hour up to 2 hours both at room temp and in a 36 degree water bath with no luck at getting the sections to stain darker. We use a 10% solution of this, so it comes out to 2% final concentration: we use 4ml of the ammonium sulfide we buy in 36ml water. I don't remember the posting but in practical use ammonium sulfide is easily added in excess of the need for the reaction, so in that sense concentration does not matter much. But maybe if you made it super low concentration it would not work as well or as fast. For our purposes 2% final concentration and up is fine. I was hoping someone might have some suggestions. I have been using clone VG1 for a while. I have never really been particularly happy with it. The labeling is not nearly as specific as I would like and there is almost always some background. If I dilute it more the signal dies off well before the background. I've been retrieving in Tris-EDTA for a half hour, diluting the primary at 1:30 and detecting with Envision (Dako) and DAB. Recently I tried a rabbit monoclonal (I usually love these) from Biocare Medical. I can certainly say it isn't as dirty as the VG1 clone, unfortunately there is not much signal at all. The recommended dilution is 1:100 to 1:200. I have tried as low as 1:50. Biocare tech support (nice guys) suggested retrieving for an hour which I tried. I have been increasing the incubation time from a half hour to 90 min to overnight in the fridge. Honestly guys, there's not much hair left to pull out. Does anyone have any suggestions? I'd really like to get this working properly. Even if it were successful spores in the current environment could still regrow. You need to sterilize the storage area. An azide soak might prove more useful as the residual azide would inhibit further growth. Azide is very toxic so this would need to be taken in account when the tissue is processed... Another idea would be to use mothballs. The 1,4-dichlorobenzene has antifungal properties. I don't know how this will effect future processing of the tissue. Bagging blocks in plastic bags with mothballs might make the storage space more user friendly. We now have 2 instruments linked together via a server with 1 PC running both. We run all the silver stains daily. GMS, WS, RETIC, PMS (jones), plus all the other regulars. Consistently good results.
