No. Re-hydrating the tissue will remove most (if not all) of the eosin and the hematoxylin will serve as your counterstain (although you may need to counterstain again at the end of you protocol). If you use antigen retrieval for an unstained slide, you will have to use it on the H&E-stained slide to uncover your target.

I would like to perform IHC (DAB) after H&E. Do I need to remove H&E? If answer is YES, my question HOW? Can I ask the protocol for that?
